چکیده

Asymmetric distribution of proteins and other molecules within cells leads to cell polarization. One of the most studied examples occurs in epithelial cells that polarize to form apical and basolateral surfaces (Nelson, 2003). Although we know much about directed protein trafficking after cells are polarized, less is known about the initial mechanisms that lead to polarization. Genetic and biochemical studies in mammalian systems and lower organisms have begun to reveal the pathways that control this process. The current model is that apical and basolateral protein complexes are mutually antagonistic, which leads to the distribution of proteins in a polarized fashion (Bilder et al., 2003; Tanentzapf and Tepass, 2003). In this model, distinct protein kinases become localized in a polarized fashion and, through phosphorylation, control the localization of other proteins (see below).

Protein complexes that control renal epithelial polarity.

Establishment of epithelial apicobasal polarity is crucial for proper kidney development and function. In recent years, there have been important advances in our understanding of the factors that mediate the initiation of apicobasal polarization. Key among these are the polarity complexes that are evolutionarily conserved from simple organisms to humans. Three of these complexes are discussed i...

VE-cadherin interacts with cell polarity protein Pals1 to regulate vascular lumen formation

Blood vessel tubulogenesis requires the formation of stable cell-to-cell contacts and the establishment of apicobasal polarity of vascular endothelial cells. Cell polarity is regulated by highly conserved cell polarity protein complexes such as the Par3-aPKC-Par6 complex and the CRB3-Pals1-PATJ complex, which are expressed by many different cell types and regulate various aspects of cell polari...

Par6b Regulates the Dynamics of Apicobasal Polarity during Development of the Stratified Xenopus Epidermis

During early vertebrate development, epithelial cells establish and maintain apicobasal polarity, failure of which can cause developmental defects or cancer metastasis. This process has been mostly studied in simple epithelia that have only one layer of cells, but is poorly understood in stratified epithelia. In this paper we address the role of the polarity protein Partitioning defective-6 hom...

Dual Roles of Notch in Regulation of Apically Restricted Mitosis and Apicobasal Polarity of Neuroepithelial Cells

How the mitosis of neuroepithelial stem cells is restricted to the apical ventricular area remains unclear. In zebrafish, the mosaic eyes(rw306) (moe/epb41l5(rw306)) mutation disrupts the interaction between the putative adaptor protein Moe and the apicobasal polarity regulator Crumbs (Crb), and impairs the maintenance of neuroepithelial apicobasal polarity. While Crb interacts directly with No...

Genes Regulating Epithelial Polarity Are Critical Suppressors of Esophageal Oncogenesis

Esophageal cancer is an aggressive disease featured by early lymphatic and hematogenous dissemination, and is the sixth leading cause of cancer-related deaths worldwide. The proper formation of apicobasal polarity is essential for normal epithelium physiology and tissue homeostasis, while loss of polarity is a hallmark of cancer development including esophageal oncogenesis. In this review, we s...
